- Phospholipid isolation from squid residues by supercritical carbon dioxide extraction
- Abstract
- Supercritical fluid extraction is an alternative to conventional separation process technique which takes the advantage of the enhanced solvent power of supercritical fluids (SCFs). The great solvent power of the supercritical fluids is due to the properties of density and viscosity, which are intermediate between gas and liquid. Moreover, solute diffusion in SCFs tends to be much higher than that of liquids. Also, it is possible to relatively fast extraction kinetics. Phospholipids were recovered from raw and supercritical carbon dioxide (SCO2) extracted squid viscera residues by various organic solvent extraction. SCO2 extraction was performed at 45 ℃ and 200 bar for removal of non polar lipid molecules. Phospholipids were extracted from raw squid viscera sample by chloroform, hexane, methanol, and ethanol and from SCO2 extracted squid viscera sample by ethanol. The pH was fixed at 5.7 for all phospholipids extraction conditions. Phospholipid classes were analyzed by HPLC equipped with evaporative light scattering detector. Phosphatidyl choline extracted by ethanol from SCO2 extracted residues was higher than that of extracted by ethanol from raw viscera. But phosphatidyl ethanolamine and phosphatidic acid were extracted higher percentage in raw squid viscera. The fatty acid compositions of ethanol extract from SCO2 extracted residues were analyzed by gas chromatography. Docosahexanoic acid was found in the highest percentage in phospholipid extract.
