- 철갑상어과의 어류에서 Somatolactin (SL)의 Complementary DNA와 genomic DNA를 분리하였다. 철갑상어 SL의 cDNA는 총 1184 base pairs (bp)로 5'untranslated region (UTR)이 56bp, open reading frame (ORF) 영역이 699 bp 그리고 3'UTR는 402 bp였다. ORF영역은 232 amino acids (aa)로, 24 aa의 signal peptide와 208 aa의 long mature peptide로 구성되어 있었다. SL의 genomic 서열은 GT / AG rule이 잘 보존되어 있었으며 4 개의 intron과 5 개의 exon으로 이루어져 있다. 유전자 발현 수준은 수정란에서부터 부화 후 11 일까지 측정되었다. 발현은 수정 초기부터 검출되어 부화 후 증가하는 경향이 있었고 난황 흡수시기와 첫 번째 외인성 먹이 섭취 후에 크게 증가했다. 철갑상어 SL mRNA 조직분포 양상을 확인한 결과 SL mRNA는 뇌 조직에서만 검출되었다. 계통 발생 분석 결과, 철갑 상어 SL은 SL- alpha 계통군과 cluster를 이루었다. 이미 다른 종에서 밝혀진 SL염기서열을 통해 철갑상어의 SL과 cysteine dependent molecular evolution model을 만들었다.
Complementary DNA and genomic sequences representing Somatolactin (SL) was isolated from Acipenseridae family fishes. The cloning of Sturgeon SL resulted the complete cDNA of 1184 base pairs (bp) that included 56 bp of 5'untranslated region (UTR), 699 bp of open reading frame (ORF), and a 3'UTR of 402 bp. Open reading frame translated into 232 amino acids including 24 aa signal peptide and 208 aa long mature peptide. Genomic sequences of SL contained five translated exons interrupted by four introns with conserved GT/AG exon/intron boundary rule. Gene expression levels were quantified starting from fertilized eggs to eleven days post hatching. Expression was detected early as fertilized eggs and tended to increase after hatching and greatly increased after yolk sack consumption and first exogenous feeding. Tissue distribution analysis showed that SL mRNAs were detected only in brain tissues. Phylogenetic analysis showed that sturgeon SL clustered with SL alpha clade. Previously published SL sequences from other species analyzed to further categorize SL and a cysteine dependent molecular evolution model was suggested.
