대황 유래 추출물을 함유하는 Polyvinyl Alcohol Hydrogel의 상처치유 효과
- Abstract
- Eisenia bicyclis is a brown edible seaweed and contains large amount of physiologically active substances such as phlorotannin. Active substances isolated from E. bicyclis exhibits various biological activities such as antibacterial, anti-inflammatory, anti-hyperlipidemia and anti-diabetic. The present study is aimed to investigate the antibacterial and anti-inflammatory effect of fraction of E. bicyclis extract and prepares polyvinyl alcohol (PVA) hydrogel containing E. bicyclis extract and analyzes its wound-healing effect in vitro and in vivo.
First, the useful components of E. bicyclis are extracted using ethanol and fractionated using organic solvents, and then antibacterial activity of each fraction analyzed by disk diffusion assay and minimum inhibitory concentration (MIC) assay for 4 types of skin pathogens. Anti-inflammatory effect is tested by nitric oxide (NO) assay. PVA hydrogel containing the ethyl acetate fraction (EB-EtOAc/PVA hydrogel), which has the highest antibacterial effect among the E. bicyclis fractions, shows no cytotoxicity, and shows a concentration-dependent NO reduction effect, is made by freezing-thawing method. The antimicrobial efficacy and cytotoxicity of EB-EtOAc/PVA hydrogel were evaluated in vitro, and the in vivo test for the wound healing effect was performed in a total of 4 experimental groups (EB-EtOAc/PVA hydrogel, PVA hydrogel, Epi-derm and untreated group) were applied to evaluate the extent of wound area recovery from day 0 to day 14. EB-EtOAc/PVA hydrogel showed antimicrobial effect in vitro, and did not show cytotoxicity. As a result of in vivo test, EB-EtOAc/PVA hydrogel treatment group showed faster recovery of wound area than untreated group, and skin irritation was reduced. It did not appear to occur, and was confirmed to have an effect on inhibiting dermatitis and healing wounds. Thus, based on the present study it is concluded that the EB-EtOAc/PVA hydrogel, which has antibacterial and anti-inflammatory effects, is effective in treating skin wounds and can suppress the occurrence of inflammation. This study opens a new door in the research industry for the application of seaweed-derived natural products especially in the cosmetics industry, formulation of skin medicines and also as a sanitary aid.
