Optimization of Rutin Extraction from the Buckwheat hulls by Response Surface Methodology
Abstract
Rutin (quercetin-3-rutinoside), one of main component has been proven to have several pharmacological properties such as antioxidant, antiplatelet, antiinflammatory activities and so on, However, optimizing extraction of rutin from Buckwheat hulls, the by-products of buckwheat, has rarely been reported. buckwheat hull is largely discarded as waste, Therefore, there has been an increased need to investigate an efficient means to utilize this by-product. This study was aimed to determine optimal extraction conditions for rutin of buckwheat hulls by using response surface methodology(RSM).
Rutin from buckwheat hulls was extracted using ethanol as a solvent with several extraction conditions. Extraction conditions were temperature (55, 60, 65℃), extraction time (2.5, 3.5, 4.5 h) and solvent concentration (60, 70, 80% v/v). 14 experiment was composed for the central composite design (CCD), six replicates were applied for zero point, and the regression model was obtained to predict the optimal extraction yield. The coefficient of determination (R2) and adjusted coefficient of determination (R2adj) for the model was 0.907 and 0.823, and the probability value (p<0.01) demonstrated a high significance for the regression model. The optimized conditions of extraction rutin from buckwheat hulls were found to be: optimized ratio of extraction temperature 59.9℃, extraction time 4.2 h and extraction concentration 72.1% of ethanol in water, and the maximum rutin yield was 354.24 mg/L. Under these conditions, validation experiments were done and the mean extraction yield of rutin was 346.22±0.67 mg/L, which is in good agreement with the predicted model value
