흰다리새우(Litopenaeus vannamei)에서 Sulfadimethoxine의 약물동태학적 및 독성 연구
- Abstract
- This study was conducted to investigate the pharmacokinetic and toxicological effects of Sulfadimethoxine (SDM), a type of sulfa drug, on whiteleg shrimp in crustaceans. The drug (SDM) was administered orally for 7 days under different conditions of water temperature (20 ℃ and 28 ℃) and drug concentration (200 and 1000 ㎎/ℓ) to the white leg shrimp. After drug administration, the amount of drug residual (muscle, hepatopancreas, and hemolymph) over time was evaluated through LC-MS/MS. The residual amount of drug was highest in all groups immediately after administration of the drug, and then gradually decreased with time. Pharmacokinetic results were derived from the drug residual results obtained using the Pk Solver program. The pharmacokinetic results of all tissues (muscle, hepatopancreas, and hemolymph) were different according to the conditions of water temperature and drug concentration. In addition, in order to evaluate the toxicity that may occur when the drug is administered to white leg shrimp, hematologic indicator analysis of hemolymph (blood count, GOT, GPT, Total Cholesterol, Glucose, Total Protein) and histopathological analysis (muscle, hepatopancreas, antennal gland) was performed. Drugs (200 and 1000 mg/ℓ) were orally administered for 7 days at an appropriate water temperature of 28 °C, and samples were collected 1 day after that. As a result of analysis of hematologic indicators of hemolymph, there was no significant difference in the drug-administered group (200 and 1000 ㎎/ℓ) compared to the control group. On the other hand, in the pathological histological analysis, neither the control group nor the drug-administered group (200 and 1000 ㎎/ℓ) showed specific necrosis and cell-destructive lesions such as inflammation.
Taking into account the above results, this study reflects various conditions such as water temperature (20 ℃, 28 ℃) and dosage concentration (200, 1000 ㎎/ℓ) that can occur when drugs are actually used in aquaculture. Therefore, it can be used as basic data on drug metabolism and toxicity that occurs when SDM is administered orally to white leg shrimp. In addition, it is believed that the results of this study can be also used as basic data necessary for drug approval/licensing, establishment of the maximum residual limit (MRL) and establishment of the period of withdrawal in the future.
