Analysis of immune-related genes in haemocytes of shrimp Fenneropenaeus chinensis infected with white spot syndrome virus by cDNA microarray

- Abstract
- cDNA microarray was utilized to identifiy differentially expressed genes in the Fenneropenaeus chinensis infected with white spot syndrome virus (WSSV). 1452 expressed sequence tag (ESTs) which were comprised of 338 contigs and 226 singlets by assembly, were discovered by the cDNA library that was constructed from total RNA of infected and non-infected shrimps' haemocytes. The immune-related genes, anti-lipopolysaccharide factor, hemocyte kazal-type proteinase inhibitor, lysozyme, profilin and protease inhibitor, were identified in the ESTs. A DNA chips was made with 403 clones that contain unique genes. The cDNA array was hybridized with labeled probes which were produced from shrimp haemocyte that was collected with a time series of 1, 2, 3 and 5 days post-injection. Seventy six differentially expressed genes were detected in which 40 were up-regulated and 36 were down-regulated. Out of the 76 genes, four immune-related genes, hemocyte kazal-type proteinase inhibitor, penaeidins 3-1, a single WAP domain-containing protein and cytochrome C oxidase subunit 1 were identified.
