폐 녹조류 재활용을 위한 Bacillus licheniformis TK3-Y균주의 분리 및 최적화

- Abstract
- Due to London dumping convection, in Korea, ocean dumping of fishery waste has been strictly prohibited since 2012 and effective reutilization of them has been being studied. Among the fishery wastes, 90 thousands tons of waste are seaweeds. For reutilization of green seaweed waste, isolation of potential microorganisms degrading polysaccharides such as, cellulose, pectin, xyloglucan and ulvan etc. was focused. Strain screening was carried out in 100-ml flask containing 1.5 g piece of green seaweed. The inoculums used in this experiment were 1 g sample taken from marshy areas near at coast, and the flask was incubated at 37℃ and 100 rpm for 21 days. After 21 days, the degradation of green seaweed was clearly seen and 0.1 ml of sample was taken from the flask, followed by spreading on agar plate containing 0.1% green-seaweed powder. After sub-culturing, some strains were newly isolated. To evaluate algal-polysaccharide degradation ability, the culture broth was spotted on agar plates containing 0.1% green-seaweed powder and 0.1% carboxymethylcellulose (CMC), and then incubated at 37 ℃ for 2, 4 days. Using Gram’s iodine solution, the clear zone was formed around the colony from one colony and identified and named it as Bacillus licheniformis TK3-Y. As a result of optimization of cultivation conditions, Bacillus licheniformis TK3-Y was well grown at pH 6 and 55℃, under CMC containing glucose and xylose as a primary substrate.
