3T3-L1 지방세포의 adipogenesis에 대한 외톨개모자반 추출물의 저해효과

Abstract
Myagropsis myagroides is a brown alga that is commonly used as animal feeds in Korea was shown to have angiotensin-converting enzyme inhibitory, hepatoprotective and anti-inflammatory activities. However, anti-obesity activitiy of Myagropsis myagroides extract has not been studied yet. In this study, 3T3-L1 cells were treated with Myagropsis myagroides ethanolic extract (MEE) or n-hexane fraction of Myagropsis myagroides (HFM) to evaluate its anti-adipogenetic activities. MEE or HFM did not show any cytotoxicixy up to 400 ug/ml in 3T3-L1 preadipocytes. When 2-day postconfluent 3T3-L1 preadipocytes were treated with MEE or HFM, lipid accumulations in cells were markedly reduced by the treatment as analyzed by Oil Red O staining. Treatment with MEE and HFM significantly decreased the expression of transcription factors related with the adipogenesis such as peroxisome proliferators-activated receptors γ (PPARγ), CCAAT/enhancer-binding proteins α (C/EBPα) and sterol regulatory element-binding protein-1 (SREBP-1) as well as their downstream proteins, including acetyl coenzyme A carboxylase α (ACCα), stearoyl-CoA desaturase-1 (SCD-1) and fatty acid synthase (FAS). MEE and HFM also decreased protein levels of PPARγ, SREBP-1, FAS and ACCα. These results indicates that MEE and HFM decrease lipid accumulation in differentiating 3T3-L1 preadipocytes via the suppression of gene expressions which are related with lipogenesis and adipogenesis.
