Precise Mechanophore Activation through DNA-Based Force Transmission Systems
- Abstract
- This study investigates the mechanochemical reactivity of ultrasound-activated polymers and DNA-based materials, elucidating their structural design principles and biomedical potential. Ultrasound serves as an effective activation modality, offering deep tissue penetration and precise spatiotemporal control, while the molecular architecture determines how acoustic energy is transduced into mechanical and chemical responses. We systematically examined how different polymer architectures influence force transmission and activation efficiency, from linear polymers with strategically embedded mechanophores to complex crosslinked networks. We further explored how the programmability of DNA enables sequence-specific reactivity, controlled scission, and precise force distribution. Through structure-based material classification and structure–activity relationship analysis, we established a comprehensive framework for designing ultrasound-responsive materials with improved performance. Building on this foundation, we utilized DNA as a versatile scaffold for polymer mechanochemistry, exploiting its inherent programmability and mechanical properties to achieve efficient ultrasound-induced molecular scission. By engineering DNA– mechanophore–DNA constructs with tunable lengths (100–1000 bp) and diverse synthetic mechanophores, we achieved nearly quantitative mechanophore activation (up to 99.9% scission within 15 minutes for constructs ≥250 bp). Selective bond cleavage was validated by DNA sequencing and mass spectrometry, demonstrating markedly enhanced efficiency and analytical resolution compared to conventional synthetic polymer systems. To quantitatively assess the effect of mechanophore positioning on activation, we evaluated the spatial reactivity of two mechanophores: disulfide bonds and site-specific nicks. Comparative mapping of their reactivity at multiple positions along the DNA backbone revealed that disulfide mechanophores exhibit higher reactivity and broader activation profiles than nick sites, in agreement with molecular dynamics simulations. Co-incorporation of both mechanophores enhanced overall reactivity while maintaining individual selectivity, establishing quantitative design principles for spatially controlled force-responsive DNA systems. These findings demonstrate that DNA-based platforms provide highly efficient and precise tools for mechanophore development and represent a versatile strategy for mechanochemical research. The proposed design principles address key challenges in activation efficiency and reaction selectivity and are expected to facilitate the development of advanced materials for diverse applications.|본 연구는 초음파 활성화 고분자 및 DNA 소재의 기계화학적 반응성을 체계적으로 분석하고, 이들의 구조 설계 원리 및 생체의학적 응용 가능성을 탐구하였다. 초음파는 깊은 조직 침투력과 정밀한 시공간적 제어가 가능한 효과적인 활성화 메커니즘을 제공하며, 분자 구조는 음향 에너지를 유용한 기계적/화학적 반응으로 변환하는 방식을 결정한다.
전략적으로 배치된 메카노포어를 포함하는 선형 고분자부터 복합 네트워크 구조에 이르기까지 다양한 고분자 구조가 힘 전달과 활성화 효율에 미치는 영향을 분석하였다. 또한, DNA의 프로그래밍 가능성이 서열 특이적 반응성, 제어된 절단, 그리고 정밀한 힘 분포를 가능하게 하는 방식을 탐구하였다. 구조적 특징에 따른 소재 분류와 구조-기능 상관관계 분석을 통해, 향상된 성능을 지닌 초음파 반응성 소재 설계를 위한 체계적인 프레임워크를 제시하였다.
이러한 이론적 기반을 바탕으로, DNA를 고분자 기계화학의 다목적 프레임워크로 활용하여 효율적인 초음파 유도 분자 절단을 달성하기 위한 힘 전달 매체로 사용하였다. 조절 가능한 길이(100~1000 bp)와 다양한 합성 메카노포어를 가진 DNA-메카노포어-DNA 구조를 설계함으로써, 초음파를 이용하여 거의 완전한 메카노포어 절단(250 bp 이상 구조체에서 15분 내 99.9% 절단)을 달성하였다. DNA 시퀀싱과 질량분석법을 통해 DNA 구조체에서의 선택적 결합 절단을 확인하였으며, 이는 기존 합성 고분자 시스템과 비교하여 획기적으로 향상된 효율성과 분석 용이성을 보여주었다.
나아가 메카노포어의 공간적 배치가 활성화에 미치는 영향을 정량적으로 규명하고자, 이황화 결합과 부위 특이적 닉이라는 두 가지 메카노포어의 위치별 반응성을 체계적으로 평가하였다. DNA를 따라 여러 위치에서 각 메카노포어의 반응성을 매핑하고 비교한 결과, 이황화 메카노포어는 닉 부위보다 높은 반응성과 더 넓은 활성화 프로파일을 보였으며, 이는 분자 시뮬레이션 결과와 일치하였다. 두 메카노포어를 함께 도입했을 때 개별 선택성을 유지하면서도 전체 반응성이 증가함을 확인하여, 힘 반응성 DNA 시스템의 공간적 설계에 대한 정량적 기반을 확립하였다.
본 연구를 통해 DNA 기반 시스템이 메카노포어 개발을 위한 매우 효율적이고 정밀한 플랫폼을 제공하며, 일반적인 기계화학 연구 수행을 위한 다목적 접근법임을 입증하였다. 이러한 설계 전략은 효율적인 활성화와 반응 특이성 달성이라는 주요 과제를 해결하여, 다양한 응용 분야를 위한 정교한 소재 개발을 촉진할 것으로 기대된다.
