Vibrio aguillarum로부터 Outer membrane vesicles (OMVs)의 분리 및 생화학적 특성
- Abstract
- Vibrio anguillarum is a gram-negative bacterium that causes vibriosis in approximately 50 different fish species. In this study, production of the outer membrane vesicles (OMVs) from V. anguillarum was shown by electron microscope. OMVs were formed from outer membrane surface of the cell and then vesicle fractions were released into environment. OMVs were isolated from culture broth of V. anguillarum by centrifugation and vesicle fractions showed spherical morphology through electron microscope. Protein profile of OMV was analyzed by SDS-PAGE and a 38kDa protein showed as the most amount of protein band in OMVs. The major protein band of 38kDa was identified as OmpU after performing MALDI-TOF MS analysis. Vesicles showed also enzymatic activity such as metalloprotease, hemolysin, and phospholipase activity. OMVs induced the production of pro-inflammatory cytokine such as TNF-α. These results suggest that membrane vesicle of V. anguillarum O1 has some role of pathogen in fish infection and influences immune response.
