- Abstract
- The in vitro effects of benzo[a]pyrene (B[a]P) on ovarian steroidogenesis of the chameleon goby, Tridentiger trigonocephalus were investigated. Maturing oocytes (0.45~0.50 mm) were incubated with 1, 10 and 100 ng/㎖ of B[a]P in the presence of exogenous precursor, [3H]-17α-hydroxyprogesterone ([3H]-17αP). Steroids were extracted from the incubated media and oocytes, the extracts were separated and identified by thin layer chromatography, high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC/MS). The major metabolites produced in vitro were estradiol-17β (E2) and estrone (E1) in the oocytes of 0.50 mm, 17α,20β-dihydroxy-4-pregnen-3-one (17α20βP), 17,20β,21-trihydroxy-4-pregnen-3-one (17α20β21P), 17α,20α-dihydroxy-4-pregnen-3-one (17α20αP) in the oocytes of 0.48 mm. B[a]P treatment (1 and 10 ng/ml) induced production of 17α20βP, 17α20β21P and 17α20αP in the oocytes of 0.48 mm. But 100 ng/ml of B[a]P inhibited production of these metabolites. In conclusion, these results suggested that lower concentration of B[a]P treatment (1 and 10 ng/ml) may induce the production of progestins on maturing oocytes of chameleon goby.
