Molecular Cloning and characterization of two B-type AST cDNAs from Pandalid shrimp (Pandalopsis japonica)
Abstract
물렁가시 붉은 새우의 신경 세포로부터 분리된 cricket 또는 B-type의 유전자를 포함한 두 개의 allatostatin 유전자를 가진 Pj-AST-BI과 Pj-AST-BII를 클로닝하였다. 이것은 각각 117, 187개의 아미노산을 가지고 있다. Allatostatin 유전자의 특징은 W(X)6Wamide를 가지고 있다. 본 논문에서 구축된 Pj-AST-BI과 Pj-AST-BII의 유전자 특징으로 WNKFQGSW amide를 가지고 있으며 이는 Marsupenaeus japonicus, Homarus americanus, Cancer borealis, Carcinus maenus 와 같은 다른 종류의 갑각류에서도 발견되었다. 계통학적 분석 결과 또한 다른 종류의 갑각류에 가까이 나타났다. Allatostatin의 발현을 알아보기 위해 End-point PCR을 행하였는데, 그 결과 brain에서 가장 높은 발현량을 보였고, X-organ/sinus-gland complex (XO/SG), thoracic ganglia (TG), abdominal ganglia (AG)에서도 발현을 확인하였다.
안병제거에 대한 유전자의 전사적 영향을 알아보기 위해 real-time PCR을 행하였고, 그 결과로 Pj-AST-BI의 경우, brain에서 안병제거 후 7일째 감소하는 발현 양상을 보였다. 본 논문의 결과로 Pj-AST-BI과 Pj-AST-BII는 양의 상관관계를 가지는 것으로 나타났으며, 두 allatostatin 유전자가 비슷한 물리적 조건에 의해 발현이 조절되는 것이라고 사료되어 진다.
Two distinct cDNAs encoding cricket type or B-type preproASTs (Pj-AST-BI and Pj-AST-BII) have been isolated from the nervous tissues of the shrimp, Pandalopsis japonica. PutativePj-AST-BI and Pj-AST-BII consisted of 117 and 187 amino acids, respectively. Proteolytic analysis predicted that seven and four mature peptides may be produced from the Pj-AST-BI and from Pj-AST-BII, respectively. All putative processed mature peptides belong to the W(X)6Wamide AST superfamily members. Among the Pj-AST-B peptides, WNKFQGSW -amide was indentified both in Pj-AST-BI and Pj-AST-BII, which were also found in the other decapod crustaceans including Marsupenaeus japonicus, Homarus americanus, Cancer borealis and Carcinus maenus. Phlylogenetic analysis showed that isolated Pj-AST-BI and Pj-AST-BII were more closely related to other crustacean AST-Bs than any other insect AST-Bs. End point RT-PCR results showed that both Pj-AST-Bs were predominantly expressed in the brain and moderate expression also found in the X-organ/sinus-gland complex (XO/SG), thoracic ganglia (TG) and abdominal ganglia (AG). In order to understand the transcriptional effects by ESA, quantitative RT-PCR was performed. Though copy number appeared to increase after 3 days of ESA but no statistical significance was identified. On the contrary, copy number of Pj-AST-BI decreased significantly in the brain after 7 days of ESA. We also found that expression of Pj-AST-BI and Pj-AST-BII is positively correlated each other suggesting their expression may be regulated in the similar physiological condition.
