Lipase inhibitory activity of Ecklonia cava extracts
Abstract
Obesity is one of the major social health problems in the world and a risk factor associated with the development of major chronic diseases, such as cardiovascular, diabetes and cancer. A fundamental cause of obesity is an energy imbalance between calorie intake and expenditure. Therefore, inhibition of digestion and absorption of dietary fat can be useful in treatment of obesity. On the other hand, high fat food is prone to lipase action, which increases free fatty acid and ultimately to the formation of off-flavors. So control of lipase is useful in food quality. This study was performed to investigate a inhibitory activity against lipase to elucidate the possible use of Ecklonia cava(EC) extracts. As a result, the lipase inhibitory activity of EC ethanol extract showed 59, 34 and 19% at concentrations of 5, 2.5 and 1 mg/mL, but water extract showed low lipase inhibitory activity. At the heat and pH stability tests, the EC ethanol extract was increased with lipase inhibitory activity by heating at 121℃ for 15 min compared with the untreated. On the other hand, the stable activity was shown under pH 2-10. Purification was carried out by liquid and liquid extraction, silica gel column chromatography and HPLC. First, the EC ethanol extract was partitioned in order of n-hexane, chloroform, ethyl acetate, butanol and water. As a result of the lipase inhibitory activity for each obtained fraction, out of four different partition layers except a water layer showed higher activity than that of the ethanol extract. Especially, the n-hexane, chloroform and ethyl acetate fractions exhibited the highest lipase inhibitory activity. The n-hexane, chloroform and ethyl acetate fractions were separated using silica gel column chromatography and were obtained 18, 15 and 22 of sub-fractions. Among them, CH1, CH2 and EA6 which showed the higher activity than those of the chloroform and ethyl acetate fractions. Especially, EA6 fraction exhibited the highest lipase inhibitory activity with an IC50 value of 1.01 mg/mL. The EA6 fraction was separated through a μBondapak C18(125 Å, 10 ㎛) column with eluted 36% ACN and 8 main peaks were obtained. Among them, a fraction 6 showed the highest lipase inhibitory activity. The results confirm the possible use of Ecklonia cava as inhibition agent of lipid degradation in food and natural anti-obesity agent.
