Abstract
Abstract

Capsosiphon fulvescens, a well-known green algae of sea algae. alge in recent years are highlighted as an anticancer drug. The present study examined whether Capsosiphon fulvescens's glycoprotein inhibits AGS human gastric cancer cell growth and mechanisms underlying its effects. AGS cells were incubated in serum-free medium with various concentrations of Cf-GP (0, 1, 2, 3 μg/ml). This study through Capsosiphon fulvescens glycoprotein (Cf-GP)'s experimental data has shown pro-apoptotic at human gastric carcinoma cells. In the results of SDS-PAGE, we confirmed that the extract from Cf-GP was a glycoprotein. Using the MTS assay and H33342 staining, we obtained Cf-GP caused cell death in a does dependent manner. MTS assay is decreased viability was induce by apoptotic cell death and H33342 staining thought the reduction of cells was observed through a microscope. In this study, effect of Cf-GP in the AGS gastric cancer was found to cell growth inhibit and IEC-6 normal cells is not toxic to cells in the proliferative effect of concentration (0, 1, 2, 3 μg/ml) was treated of 24 h to process the experiment was conducted. cell viability when a 3 ug/ml concentration in AGS cells. While, IEC-6 cells viability was compared with the control cells no significant change. Also, we observed changes through western blotting in the Fas signaling and mitochondria pathway. To determine effect of Cf-GP on apoptosis events, cells were treated Cf-GP and examined expression of several proteins related with apoptosis by western blotting. In the result of western blot, we obtained Cf-GP activated caspase-cascade and PARP, which is a substrate of caspase-3, -8 and proteins expression of bcl-2 family. In addition we have experiment of caspase-3, -8 activation and annexin V. Also we observed cell cycle test assay and expression of cell cycle related proteins. This result through where arrest has occurred and there has increased sub-G1 percent and decrease in expression of cyclin D, E, Cdk2, 4, 6 and increase in protein levels of p21, 27. In This study demonstrate that Cf-GP triggers apoptosis of AGS cells through activation of the caspase pathway. Moreover, expression of Bcl-2 family proteins may play an important role in Cf-GP induced apoptosis. In conclusion, the results of the present study indicate that Cf-GP inhibits proliferation and induces sub-G1 phase arrest and apoptosis in AGS human gastric cancer. So we make certain apoptosis at gastric cancer. Therefore, this study has suggest that Cf-GP could be a potential source of bio-functional material to have anticancer effect in human gastrointestinal cancer and Cf-GP presents potentially interesting antitumor drug and food.
