Proteomic 분석을 통한 Vibrio anguillarum의 스트레스 대응 단백질에 관한 연구

- Vibrio anguillarum은 호염성, gram-negative의 대표적인 어류병원성 수해양 미생물로 인체 및 어류에 큰 피해를 주고 있다. 이러한 V. anguillarum을 다양한 환경적 stress 하에 둠으로써 이에 따른 단백질의 발현을 2-demensional electrophoresis 와 matrix-assisted laser desorption ionization-time of flight/mass spectrometry (MALDI-TOF/MS)를 통해 분석하였다. 그 결과 wild type의 경우 550여 개의 spot이 나타났으며, 15℃의 온도 stress를 받은 균의 경우 spot의 양이 530여 개로 감소한 반면 pH stress를 받은 균의 경우 총 580여 개로 증가하였다. 이들 spot 중 40개의 spot이 identification 되었다. 온도 stress의 경우 aspartyl-tRNA synthetase, ABC transporter ATP-binding protein을 비롯하여 11개의 spot이 up-regulation되었으며 heat shock protein 9, putative C4-dicarboxylate transport sensor protein 등 6개의 spot이 down-regulation 되었다. pH stress의 경우에는 general secretion pathway protein D precursor, adenylyltransferase 등 23개의 protein이 up-regulation 되었으며 Ribosomal protein S2, ADP-L-glycero-D-mannoheptose-6-epimerase 등 18개의 protein이 down-regulation 되었다. oxidoreductase short-chain dehydrogenase/reductases family protein (OSDR) 의 경우 온도와 pH stress를 비롯해 NaCl 농도 (2M), Bile salt 농도 (0.1%)의 변화에도 protein이 down regulation 되었다. Stress에 민감하게 반응하는 OSDR의 DNA sequencing을 통해 759 bp의 ORF를 밝혔으며, 이를 바탕으로 252 amino acid 서열을 찾았다. 또한 OSDR protein의 발현의 조절 mechanism을 찾고자 RT-PCR을 실시한 결과 stress 조건 하에서 OSDR은 시간이 지남에 따라 급격히 감소하였고, 이를 통해 OSDR protein의 발현의 조절이 transcriptional level에서 일어남을 알 수 있었다. 본 연구를 통해 V. anguillarum의 OSDR이 다양한 환경적 stress에 대해 매우 sensitive하게 작동함을 밝혔으며 또한 이러한 결과를 바탕으로 V. anguillarum의 stress 여부를 측정하는 satellite maker로 OSDR이 적합하다는 결론을 도출하였다.
Vibrio anguillarum is a halopilic and gram-negative bacterium that universal marin pathogen causing vibriosis in marin fish and other aquatic animals. In general, bacteria have developed strategies to survive in harsh environments. V. anguillarum also encounter various stresses with seawater and hosts. In this study, we investigated differential protein expression of the V. anguillarum in response to pH and temperature stresses. V. anguillarum was cultured under stress condition A (15 ℃, pH 7), stress condition B (25 ℃, pH 10), and normal condition (25 ℃, pH 7), respectively. Proteins were separated by 2D-PAGE, and these proteins were observed different expression in each conditions, and identified by MALDI-TOF/MS. About 550 proteins in normal condition, about 530 proteins in stress condition A, and 580 proteins in stress condition B were expressed on the gel. Among them, 40 proteins were indentified, and 2 proteins were commonly down-regulated under some stress conditions. This proteins were identified as putative C4-dicarboxylate transport sensor protein and oxidoreductase short-chain dehydrogenase/reductases family protein (OSDR). Especially, OSDR was down-regulated at pH (5), NaCl concentation (2M), and bile salt (0.1%). OSDR was included in short-chain dehydrogenase/reductases (SDR), and it has distinct sequence motifs as TGXXXGXG. Analysis of transcriptional level with RT-PCR showed that OSDR gene was reduced by time course. As a result, expression of OSDR protein was regulated in transcriptional level. To the best of our knowledge, this is the first report describing stress-responsive proteins of V. anguillarum using proteomic approaches. Our results provide a useful strategy for detection of changed environment with V. anguillarum.
